Before entering politics, Agboola Ajayi held positions as a lawyer and businessman and was associated with various renowned and international corporations.
His political journey began in 1999 when he served as the Director of Agriculture in Ese-Odo Local Government Area and later as the Director of Labor and Transportation in the same area until 2003.
He then became the Chairman of the Caretaker of Ese-Odo Local Government Area The government from 2003 to 2004 was then successfully chaired, which he held from 2004 to 2007. During this period, he became the Local Government Association of Nigeria (ALGON); also treasurer. In 2007, Ajayi was elected to represent Ilaje/Ese-Odo federal constituency in the Federal House of Representatives and served as the Chairman of the House Committee on NDDC from 2007 to 2010.
Between 2007 and 2010, Hon. Alfred Agboola Ajayi served in the NDDC and was on various committees, including the House Committee on Justice, Housing, Industries, and Gas. In 2016, he joined the APC and became the electoral partner of Arakunrin Oluwarotimi Akeredolu, SAN, winning the election. Both were formed on February 24, 2017.
On June 24, 2020, Ajayi switched to PDP and declared his intention to challenge Akeredolu in the gubernatorial election. The Daily Post reported on July 6, 2020, on plans to remove Ajayi from office due to tensions between him and Akeredolu, with the process likely to commence at the Ondo State House of Assembly.
